These cannot be doubled up — if you have an Australian degree, completing the PY doesn't add another 5 points on top. The PY points apply to those who did not complete their degree in Australia but complete a PY after arriving.
Additionally, for ACS-assessed occupations, completing the Professional Year effectively eliminates the experience deduction for that period — the PY itself counts as skilled employment at the required level.
